| A potent inhibitor of Wnt signaling (EC50 of ~10 nM). This was recently described as an activator of CK-1alpha that subsequently blocked beta-catenin signaling. The paper shows that pyrvinium binds all casein kinase 1 (CK1) family members in vitro at low nanomolar concentrations and pyrvinium selectively potentiates casein kinase 1a (CK1a) kinase activity. Thus representing a more potent inhibitor of the Wnt/betacatenin signaling pathway than other compounds currently available. See (Nat Chem Biol. 2010 Nov;6(11):829-36). |
